Product description

ZORIA-005 is a calcium-enriched variant of the selective supplement, providing stronger suppression of bacterial growth while preserving the viability of yeasts and fungi. Intended for mycological media — chiefly Sabouraud and its derivatives — where you want to limit accompanying bacterial flora without reaching for chloramphenicol, ampicillin or other antibiotics.

This matters wherever an antibiotic in the medium gets in the way: it raises cost, is unstable, complicates waste disposal and imposes selective pressure toward resistance. ZORIA-005 solves the problem differently — the active substance is ascorbic acid and its salts in a proprietary composition with magnesium and calcium ions (patent application P.454716). A bacteriostatic, non-antibiotic mechanism, so no risk of selecting resistant strains.

Compatibility limit: because of its calcium content the product is not intended for phosphate-buffered media — calcium ions form a poorly soluble precipitate with phosphates (turbidity, cells bound in the precipitated phase). Compatible media: Sabouraud and derivatives (peptones + glucose, phosphate-free). Incompatible: TSB, LB and other phosphate media — for those use the calcium-free ZORIA-001.

Preparation

  1. Dissolve the sachet contents in 150 ml of distilled or molecular-biology-grade water.
  2. Filter the solution through a 0.22 µm filter.
  3. Add to 850 ml of target medium at a temperature below 60 °C.
The additive may be dissolved directly in the target medium; note, however, that medium components may clog the filter faster.

Scope of research applications

  • Research selective cultivation of yeasts and filamentous fungi with suppression of accompanying bacterial flora.
  • Work with species sensitive to classic antimicrobial agents (chloramphenicol, cycloheximide).
  • Mycological media (e.g., Sabouraud) where antibiotic pressure should be avoided.

Technical notes

  • The active substance is heat-labile. Do not autoclave, boil or microwave solutions containing the additive. Add the working solution only to media cooled below 60 °C.
  • The active substance is sensitive to light and oxygen. Store solutions protected from light, in containers filled as close to the cap as possible.
  • A colour change to orange or brown indicates degradation of the active substance. Medium with a discoloured additive should be discarded.
  • For spectrophotometric measurements, run a dynamic blank for each condition.
  • The additive is compatible with semi-solid and solid agar-containing media.
  • Compatibility restriction: Product is not intended for media containing phosphate buffers. Calcium ions form a poorly soluble precipitate with phosphates, causing medium turbidity and cell binding in the precipitate phase.
  • Compatible media: Sabouraud and derivatives (peptones + glucose, without phosphates). Incompatible media: TSB, LB with phosphate additives, phosphate-buffered media.
  • Calcium may affect cell aggregation — for quantitative measurements, microscopic assessment of the suspension before plating is recommended.
  • The additive can cause a drop in pH by 1-2 units.

Safety note

The active substance is not classified as hazardous under the CLP Regulation. Calcium chloride is not classified as hazardous in the concentrations used; concentrated solutions may be irritating to the eyes.

  • Ingestion: rinse the mouth with water.
  • Eye contact: flush with plenty of water or saline.
  • Skin contact: the working solution has a low pH and may irritate mucous membranes and damaged or inflamed skin. Healthy skin — rinse with water.
Disposal of small quantities: dilute with plenty of water. Dispose of larger volumes in accordance with applicable waste regulations.
